By 2018, Detroit has become devastated by countless criminal activities and violence. The authorities constructed a large wall to house criminals in a community known as the Brick Mansions. The Mayor (Bruce Ramsay) and his associates plan to make the city prosper, with the intention to do something drastic about Brick Mansions.

Two gangsters, K2 (Gouchy Boy) and Rayzah (Ayisha Issa), kill two guards outside one of the mansions to find Lino Dupree (David Belle) since he stole 20 kilos of cocaine from their boss, the drug kingpin Tremaine Alexander (RZA). The gangsters storm the place as Lino disposes of the drugs. He runs and evades the crooks. They chase after him and try cutting him off, but he fights them off. Lino hops to the roofs, still running. A gangster throws a grenade at him, and Lino jumps off the roof onto another building to safety.

We meet police officer Damien Collier (Paul Walker), who has his own agenda against Tremaine because he killed Damien's father. He meets with wanted mobster George the Greek (Carlo Rota) in a restaurant. George takes Damien and other associates to his drug house where all his products are being put together. The mobster realizes he's been bugged, giving Damien the chance to arrest him. George's goons start shooting, giving him a chance to run. Damien escapes the gunfire and chases after the getaway car. He jumps onto it and breaks in, crashing the car into the police station so that George is launched from his seat right into the station so that he can be booked.

K2 and Rayzah report back to Tremaine as he's cutting peppers with a huge cleaver. He holds it to K2's throat for his failure to get Lino. K2 tells Tremaine that Lino has a girlfriend (really his ex), and if they get her, Lino will go to them. Tremaine lets them leave, but tells them not to return without her. The girl, Lola (Catalina Denis), is working at a diner when Rayzah shows up and pulls a weapon on her. Lola runs but is knocked out cold by K2. They grab her and throw her in their car.

Damien visits his grandfather (Frank Fontaine), who wishes he would not pursue Tremaine, as it will not do any justice to seek revenge.

The goons return with Lola, preparing for Lino to come back for her. Sure enough, Lino crashes through the ceiling. He holds Tremaine at gunpoint, and both he and Lola escape with Tremaine. They drive away, with Tremaine's goons following. Lino and Lola take Tremaine into a prison, but it turns out that one of the guards works for Tremaine. Lino is locked up and Lola is kidnapped again. After Tremaine leaves, Lino grabs the guard's head and places it through a slot in the door, allowing Lino to stomp the guy's head and break his neck.

Damien is brought in to meet with the Mayor after Tremaine and his goons hijacked a truck carrying a powerful neutron bomb, and that it has been triggered. The mission is for Damien to infiltrate Brick Mansions and contact the Mayor and company for a code to deactivate the bomb. He has ten hours to do so, and they pair Damien up with Lino as his eyes.

Posing as a prisoner, Damien is taken to the Michigan State Penitentiary where Lino is held. The two of them are transported in a van where Damien picks the lock on his handcuffs and frees himself and Lino. Damien kicks both guards out of the van and drives away. He and Lino get into a fight as they both try to take over the van to get back to Brick Mansions, but Damien succeeds in driving away. Lino hops in through the window into the passenger's seat. The cops come after them, forcing Damien to drive past a checkpoint around Brick Mansions where they crash into a lot full of cars. Lino handcuffs Damien to the wheel and leaves him. Some thugs approach Damien since the car he crashed into belongs to one of them. Damien breaks the wheel off and fights the thugs, managing to get away again.

Damien finds Lino since he put a bug in his pocket during their fight. Damien convinces Lino that he does care about what happens in Brick Mansions, so they agree to work together to confront Tremaine themselves. Lino calls Tremaine on Damien's phone and tells him they're coming.

When left alone, Rayzah messes with Lola. They get into a fight, with Rayzah viciously beating Lola down. She nearly kills her until Tremaine contacts her through a walkie-talkie, telling her to take it easy.

Damien and Lino are escorted up to Tremaine's place. He agrees to disable the bomb for a price. If he gets ten dollars from the 3 million residents of Brick Mansions, he can get $30 million. Damien contacts the authorities to make a negotiation, but that kind of money can't be found in Brick Mansions. Damien bluffs the deal and says it's a go. He and Lino are kept as prisoners until Tremaine gets his money. Damien admits the bluff to Lino and also tells him about Tremaine killing his father. Lino empathizes with Damien and begins to fake a choking fit to fool the guard into letting them out.

Tremaine watches as Rayzah keeps her eye on Lola next to the rocket that holds the bomb. He admits to K2 that he has uneasy feelings over this.

Once again, Damien and Lino outrun and fight off more goons through the area, and then get chased until they make it closer to Tremaine's place. They take down a sniper and steal his rifle. Damien shoots at Rayzah to get her away from Lola. They jump back into the building and fight off Tremaine's largest goon, Yeti (Robert Maillet). Damien ties him up and Lino kills him with a cinder block to the head. Tremaine, now realizing that his money isn't coming, grabs his gun to finish off Damien and Lino. He rounds up his goons to get ready. 

Tremaine goes up to the rocket and finally reconsiders his plan. He considers himself a businessman and refuses to kill off a whole community for his money. Rayzah tells him he's gone soft and she pulls her gun on him. He reaches for his and gets shot in the arm. Lola pounces on Rayzah and flips her over the ledge, holding onto her with her thighs. Though Rayzah pleads for her life, Lola releases her grip and lets Rayzah fall to her death.

Damien and Lino make it up to the rocket. Damien requests the code from the Mayor and company, which turns out to be the same zip code as Brick Mansions. This causes Lino to have doubts. He fights with Damien after they argue over entering the code. As the time continues to run out, Damien holds Tremaine at gunpoint, now able to confront his father's killer. However, Tremaine tells him that he did NOT kill his father. He was, in fact, set up by his partners and executed, with the blame pinned on Tremaine. When the countdown on the bomb reaches zero, it is surprisingly deactivated. Damien realizes that Tremaine isn't a really bad guy, and they team up to after the real bad guys.

With a whole army of thugs, Damien, Lino, and Tremaine storm the Mayor's office with the bomb. They place it on his desk and get ready to enter the code they were sent. Before they can hit the last number, the Mayor stops them and admits that he had the bomb sent into Brick Mansions, as well as indirectly admitting his part in Damien's father's murder. This is all recorded from a camera in the distance.

The Mayor is reported to have been arrested after the video of his confession went viral. A while later, Damien drives through the Brick Mansions community, greeted by everybody he passes. Support in the community has soared, and the acting mayor has agreed to tear down the walls that separates Brick Mansions from the rest of the city. He stops by Lino's place. He is back with Lola, and the three of them support Tremaine, as he has given up his criminal ways and has decided to run for mayor. Damien and Lino part as friends.

The film ends with Damien reuniting with his grandfather, who now realizes that his grandson has found peace. There is also a brief text that says "In Loving Memory of Paul Walker", along with a picture of the man himself.
